讲座摘要:

The biological effects and biomedical application of high voltage pulsed electric fields (PEF) has become the hot spot in recent years, which has been used for sterilization, gene electrotransfection, electrochemotherapy, electrodynamic therapy and tissue ablation. Irreversible electroporation (IRE), which is based on the biological effect of PEF, is an emerging non-thermal tumor ablation technique. It uses ultrashort high-voltage electric pulses to induce cell death through apoptosis or necrosis induced by either disrupting the balance of the transmembrane potential. This causes permanent membrane lysis, or secondary breakdown of the membrane due to abundant transmembrane transfer of electrolytes and adenosine triphosphate, leading to loss of cell homeostasis. Therefore, IRE has been treated as a focal ablative technique used for certain solid tumors that are unsuitable for surgery or thermal ablation because of their precarious anatomic location. In contrast to thermal ablation, IRE can achieve tumor ablation without affecting acellular tissue constituents, such as extracellular macromolecules, connective tissue and tissue scaffold, thereby avoiding irreversible thermal tissue damage. As a consequence, these characteristics theoretically make it attractive option for tumors in the vicinity of vital structures or those located in a cavity organ. Our team has gathered medical and engineering researchers, and has done a lot of work on the principle of PEF biological effect, the research and development of new ablation equipment and catheter-based electrodes. In this talk, I will present the research progress and future prospect for high voltage electroporation pulses and its clinical applications
